You don't run your ram at the rated frequency, that is for overclocking headroom as I mentioned earlier. Go with 1333 at least (1600 is better)

So what do you actually recommend for Premier and After Effects CS5? Will I benefit more from...
DDR3 1333 12gb with a triple channel kit OR DDR 1600 6gbs triple channel kit? I've been reading around and I know that games don't need much ram.. 12gbs would be overkill and 6gbs is more than enough. How much ram would I need for after effects and premiere to run smoothly? Will I even see a difference between the two configurations?

12GB DDR3 1600 kit for $300 either one is good and I bet you can find people running these kits fine in that board.

The drives are limited in theri throughput anyway, so I wouldn't worry about it.
