using clone to remove dirt

I am editing my 24p footage with FCP 4 on my brother's mac. I need to get a couple files to my PC for dirt removal with AE. what's the best way to get the files transferred there and back in lossless format?
Thusly, what is the easiest way to use the Clone tool to remove some dirt/water spots on the lens from the frame?

Export in a PNG or TIFF image sequence. I've never used the clone tool in AE, but I would imagine it's somewhat similar to photoshop->use soft brush, sample often, preview zoomed in and out. I've been using Shake for all my paint work and these techniques work great for me.
