General Error-please help.

I have been editing a wedding video for a friend. I turned off the audio on the time line and continued to work. Later I shut down the computer and now everytime I try to enable the audio on the timeline and play it back I get a general error message. The same with backup video.It will play the video without the audio. Any Ideas?

I finaly got an error relating to not enough memory. I rebooted and it works now. I never use the sleep setting and the other day I started using it. It seems my mac woke up and didn't have use of all the memory. Works now.
