I'm responding to a job listing for videographers which seems to fit me perfectly (a student entry-level job for 2-3 minute productions)
They're requesting a "sample video" via email.
I have no idea what's the best and most professional looking method to submit my video.
Please help quickly!

You mean format ala , DVD, VHS, Mini DV, Beta, etc... etc... etc...
Or style for your demo reel?
well, both format and application.
Ex: should I send it as an attachment? quicktime .mov? Google video? MySpace video? etc...

Ask them. Myspace video, google video, youtube are all OUT OF THE QUESTION. Hosting it somewhere as a very decent resolution with Great compression would be good as a backup for instantaneous viewing. Use H.264 at 640 x 480 or its widescreen counter part. But those youtube style videos are for kids and arent professional at all.
But you really should ask whoever you are looking into working for what their desired format is.
As far as the demo reel itself, you should give them what is applicable to the job description and then a little taste of the extra stuff you know you can do with skill. Add some oomph so you can grow in the company. You may not get a chance to say show some editing skills for types of stuff they do.
Example, if they do weddings, show some videography stuff you have done, with nice transitions and some steady non shaky shots and good framing, and also put some graphics in there, and organize the demo reel, like start with music video editing, then weddings, and then movies, theyll see you can do more than weddings that way, and also get what they wanted to see.
But you should ask them, its not out of the question to ask a few questions about the job and what they would like to see.
