Removing FAT32 from ext. hard drive

I recently bought a new WD EXT hard drive - 600gig. The file system - FAT32 - only allows me to capture 3.99gig at a time...which is big pain in booty. Does anyone know how I can remove the FAT32 file system? I'm operating on Vista...Any help would be much appreciated.
